We conclude that mammalian Mon1, Ccz1 and C18orf8 form a stable trimeric complex, the MCC complex, of which C18orf8 is a core component. The Mon1-Ccz1-C18orf8 complex is responsible for mammalian Rab7 activation. The trimeric Mon1-Ccz1-C18orf8 (MCC) complex therefore acts as an activating GEF for mammalian Rab7 and its role in late endosomal trafficking and LDL-cholesterol uptake can be specifically by-passed by a constitutively active Rab7 (Q67L) or by Rab7GAP depletion. |
