Description MLL-AF4 fusion protein is the product of the t(4;11)(q21;q23) chromosomal translocation, and it is the most frequent genetic aberration of the human MLL gene, resulting in high-risk acute lymphoblastic leukemia. This oncoprotein is unfortunately associated with poor prognosis and typical manifestations such as a large number of white blood cells. Generally the AF4 gene encodes a protein with serine- and proline-rich regions located near the AF4 breakpoint and a nuclear localization signal domain located to the breakpoint. On the other hand MLL gene contains two central zinc-fingers, a C-terminal region with homology to Drosophila trithorax gene, three N-terminal A/T hook regions (this kind of domain has been shown to mediate binding of MLL to cruciform DNA), a domain with homology to methyltransferase, and a proline-rich region. MLL-AF4 by itself is not sufficient to induce leukemogenesis. So the translocation between MLL and AF4 may alter expression of genes regulated by MLL or AF4.
