Description: Myostatin regulates muscle differentiation via SMAD2 and SMAD3 while follistatin counterbalances this negative modulation. Myostatin binds its receptor ActRIIB. Activated receptor phosphorylates SMAD2 or SMAD3. Phosphorylated SMADs form a heterocomplex with SMAD4 and translocate into the nucleus where they interfere with MYOD1 transcriptional activity and expression. This inhibition results in myogenic differentiation failure.
