The endothelial cells secrete MK, which acts on smooth muscle cells and induces production of factors including IL8. IL8 then acts on the endothelial cells and promotes their growth. Thus, MK plays a key role in the interaction between epithelial cells and mesenchymal cells. |

Midkine, also known as neurite growth-promoting factor 2 (NEGF2), plays an important role in cell proliferation, apoptosis and differentiation. Recent studies have shown that Midkine is up-regulated in several types of human cancers. In the present study, we found that Wnt3a administration or transfection of a constitutively activated β-catenin promoted Midkine expression in glioma cells. We further identified a TCF/LEF binding site, with which beta-catenin interacts, on the proximal promoter region of Midkine gene, by luciferase reporter and chromatin immunoprecipitation assays. Thus, our results suggest a previously unknown Wnt/β-catenin/Midkine molecular network controlling glioma development.β-catenin binding to the promoter region of Midkine |

Midkine is a heparin-binding cytokine or a growth factor with a molecular weight of 13 kDa. Midkine binds to oversulfated structures in heparan sulfate and chondroitin sulfate. The midkine receptor is a molecular complex containing proteoglycans. Midkine promotes migration, survival and other activities of target cells. Midkine has about 50% sequence identity with pleiotrophin. |

Syndecans are transmembrane HSPGs that act as co-receptors for MDK, facilitating clustering with other signaling partners such as LRP1 or integrins. Syndecan-1 is critical for cell–matrix interaction and angiogenesis, and in the TME, the MDK–SDC1 complex enhances fibroblast activation, ECM deposition, and metastatic invasion |

PTPζ, also known as RPTPβ/ζ, is a chondroitin sulfate-modified transmembrane phosphatase prominently expressed in the central nervous system and gliomas. MDK binding to PTPζ inhibits its phosphatase activity, leading to hyperphosphorylation of downstream oncogenic proteins (e.g., β-catenin, Fyn), which promotes cell survival and migration. This mechanism is particularly relevant in GBM and neural crest-derived tumors |
