Description: The non-canonical WNT ligand, WNT5A, binds frizzled (FZD2) receptor, hence activating the histone methyltransferase SET domain bifurcated 1 (SETDB1). SETDB1 forms a complex with chromodomain helicase DNA-binding 7 (CHD7) and NEMO-like kinase (NLK) to inhibit the ability of PPARγ to transcriptionally activate its downstream metabolic target genes. On the other hand, the canonical WNT ligand, WNT10B, inhibits adipocyte differentiation by leading off β-catenin nuclear translocation.
