Description: IL1-type cytokines are major mediators of innate immune responses, and the blockade of the founding members IL-1alpha or IL-1beta by the interleukin-1 receptor antagonist (IL1RA) demonstrated a central role of IL1 in a number of human autoinflammatory diseases. IL-1alpha or IL-1beta rapidly increase mRNA expression of hundreds of genes in multiple different cell types. In response to the ligand, a complex sequence of combinatorial phosphorylation and ubiquitination events results in the activation of nuclear factor kappaB signaling which induces the expression of canonical IL1 target genes. IL6 can be transcribed via several transcription factors. NF-kB, activated by IL1 pathway, can bind the promoter of IL6. ATF2 transcription factor also, phosphorylated by JNK, can bind the IL6 promoter enhancing its transcription.
